Production of glucosyltransferase B and glucans by Streptococcus mutans strains isolated from caries-free individuals.

چکیده
UNLABELLED Glucosyltransferase B is an enzyme produced by Streptococcus mutans, which catalyzes synthesis from sucrose of insoluble glucans that provide support to the biofilm. It is one of the main virulence factors in the generation of dental caries. However, its role is unclear in caries-free individuals who carry the bacteria. The aim of this study was to determine the production of glucosyltransferase B and the production of glucans by Streptococcus mutans strains isolated from biofilms of 30 caries-free individuals. Strains were grown in Todd Hewitt broth. Extracellular proteins were obtained by ammonium sulfate precipitation and associated membrane proteins by extraction with urea. The presence of GtfB was determined by molecular weight by SDS-PAGE and confirmed by Western blotting using a specific monoclonal antibody and polysaccharide production by electrophoretic separation, incubation with sucrose and Schiff staining. The results show that 96.7% of strains of Streptococcus mutans produce a band at the position of the molecular weight corresponding to the glucosyltransferases, of which 63.4% are positive by Western blot. Polysaccharides are produced by 93.3% of the strains. CONCLUSIONS Streptococcus mutans strains isolated from biofilm of healthy individuals produce virulence factors associated with dental caries, such as glucans and glucosyltransferase B. This indicates that there are conditions in the oral cavity different from these factors that keep the individual free from dental caries, which should be investigated in the search for strategies to control the disease.

Sucrose-dependent cell adherence and cariogenicity of serotype c Streptococcus mutans.
Four strains of serotype c Streptococcus mutans differing in glucosyltransferase (GTase) and fructosyltransferase (FTase) activities were examined. These strains had been made resistant to streptomycin. FTase activity of an S. mutans clinical variant, MT6801R, which forms large mucoid colonies on sucrose-containing agar, was considerably higher than that of a typical serotype c strain, MT8148R,...
متن کاملCharacterization of an anti-glucosyltransferase serum specific for insoluble glucan synthesis by Streptococcus mutans.
An anti-glucosyltransferase serum, which synthesized 96% insoluble glucans, was prepared against a purified enzyme preparation from Streptococcus mutans strain HS6 (serotype a). This serum was examined for its effects on glucan synthesis by crude enzyme preparations from eight strains (four serotypes) of S. mutans and for the ability of these preparations to promote adherence of S. mutans to a ...

Strains of Streptococcus ferus isolated from the oral cavities of wild rodents inhabiting sucrose-rich and sucrose-poor environments have many traits in common with the "mutans" streptococci. Thus, S. ferus HD3 and 8S1, like cariogenic S. sobrinus 6715-13, from adherent, alpha (1 leads to 3) glucopyranosyl-glucose linkage-rich, plaquelike deposits in vitro and in vivo through the action of cons...
متن کاملCariogenicity of Streptococcus mutans Glucan-Binding Protein Deletion Mutants
Streptococcus mutans is a principal etiologic agent in the development of dental caries due to its exceptional aciduric and acidogenic properties, and its ability to adhere and accumulate in large numbers on tooth surfaces in the presence of sucrose. Sucrose-dependent adherence is mediated by glucans, polymers of glucose synthesized from sucrose by glucosyltransferase (Gtf) enzymes. S. mutans m...
